中文摘要: 本项目面向心力衰竭早期检测的需求,研究基于石墨烯场效应晶体管生物传感器的心衰早期检测技术和方法,旨在纳米生物传感技术在临床疾病诊断研究和应用领域取得突破。采用传感器沟道表面功能化和机械剥离法相结合的方法共价键合单层石墨烯在传感器表面,获得可控制备基于石墨烯场效应晶体管生物传感器的新技术,构建制备简单、重复性好、价格低廉的新型纳米生物传感器。研究蛋白质分子在石墨烯表面功能化新方法,优化蛋白质与蛋白质相互作用的条件,建立基于石墨烯场效应晶体管生物传感器在血清中高灵敏度、同时检测两种蛋白质的新方法。实施该传感器对心衰病患血液样本中NT-proBNP和BNP的实时监测,探讨这两种标志物与心衰的相关性,为早期检测心衰提供新途径。本项目的建立将有利于心衰的早发现、早治疗,延长患者的生存期,并极大地减轻患者和医疗卫生系统的医疗负担,为人民健康、国民经济建设和社会和谐发展作出重要贡献。

英文摘要: Driven by the needs for monitoring heart failure (HF) in its early stage, the proposal is directed to a method of graphene-based field effect transistor (FET) biosensor for detection of HF-related biomarkers, aimed at developing a new nano-biosensing technology in the applications of disease diagnostics. In order to make simple, reproducible and cheap biosensor, a single-layer graphene can covalently be bound to the sensor channel surface by combining functionalization of the channel surface with mechanical exfoliation, pointing to a new technique for producing the new nano-biosensor in a controllable manner. The new method of highly sensitive and simultaneous detection of two proteins in serum using the graphene-based FET biosensor will be established by investigating new functionalization protocols for immobilization of protein molecules on graphene surface, and optimizing the conditions for protein-protein interactions. The biosensor will finally be used to real-time detect NT-proBNP and BNP in HF patient blood samples, which will pave a way for HF early detection. Based on the results, the relationship between HF and the two biomarkers will also be discussed.
