中文摘要: 针对当前常用的药物载体存在包封率/负载量低及药物突释等问题,考虑到金属-有机骨架材料(MOFs)具有高比表面积、孔道尺寸与形状可调和较好的生物药理安全性等优点,本项目通过预设计的金属簇来模拟硅氧四面体,定向合成系列生理安全的簇基类沸石型MOFs材料,利用功能配体的侧链基团修饰骨架材料内表面以提供特定的化学环境;在系统研究材料的合成条件、结构特征和物化性质的基础上,以半边旗中的5F及其异构体药物的负载-控释为观测平台,探究客体药物分子在主体骨架材料孔洞内的存在形式以及它们之间的相互作用方式,结合理论模拟,阐明骨架材料的药物负载-控释机理,总结其中规律,为开发新型高效的药物载体提供科学依据和实验基础,促进化学、材料、药学的交叉融合,具有重要的学术价值和实际意义。

英文摘要: In view of the current commonly problem of low closure rate/load and burst release for drug carriers, and considering the metal-organic framework (MOF) materials have high surface, well controllable core size, shape and good biological pharmacology etc., the project will be focused on the directional synthesis a series of physiological safety MOFs through the design of metal cluster to simulate silica tetrahedron. The side skeletons of functional ligands are employed to modify the material surface, which could provide certain chemical environment. Based on the drug load-controlled release of pteris semipinnala L and the system of synthetic conditions, structural feature and chemical properties of MOFs materials, the form of guest drug molecules and the interactions between them in the core will be explored. We will clarify the drug load-controlled release of mechanism and conclude the results, which will provide scientific and experiment basis for the development of new and high efficient drug carriers. This project could promote interdisciplinary relation of chemistry, material and pharmacy and has important academic value and practical application.
