中文摘要: 核酸与蛋白质的高灵敏检测在临床医学、生物工程和司法鉴定等方面具有重要意义。信号放大技术是目前提高检测灵敏度的常用方法。现有的信号放大技术虽极大地提高了生物分子检测的灵敏度,但是往往需要较长的检测时间作为代价。这种高灵敏度与高检测时间成本的矛盾,使其难以广泛应用于实际的临床检测之中。针对这一问题,本项目拟从以下几个方面开展研究:利用直链的杂交链式反应制备HCR产物,这种直链的HCR产物可通过两条核酸链直接制备,制备过程简单容易;将这种HCR产物与磁分离技术结合,通过荧光分析法和显色法即可用于特定的核酸检测。由于本方法中的HCR产物可预先制备,因而在检测时没有信号放大过程,可实现目标物的高灵敏快速检测。改变磁珠上的核酸序列,即可用于其它核酸、蛋白质及小分子的检测。

英文摘要: The highly sensitive detection of nucleic acids and proteins is of great importance in clinical medicine, bioengineering and judicial expertise. The main method for improving sensitivity is signal amplification technology. While the high sensitive is often achieved at the expense of considerable detection time with the exist signal amplification technology. This contradiction between high sensitive and high time cost make it hardly applied in clinical detection. To address these problems, this project plans to carry out research in the following aspects. The HCR products can be simply and directly prepared by straight-chain hybridization chain reaction (HCR) with two nucleic acid chains. Combined this kind of HCR products with magnetic beads separation technique, the target DNA can be determined by fluorescence and colorimetric methods. Since the HCR products can be prepared ahead, there is no amplification reaction during the detection process, and the target DNA can be determined quickly and high sensitively with signal amplification. If the sequence of the surface of the magnetic beads is changed, it can also be applied to detect other nucleic acids, proteins and small molecules.
