中文摘要: 流感病毒是重要的人畜共患传染病病原,目前应用的抗流感病毒药物在使用过程中易于导致耐药性毒株的产生。因此,研究流感病毒复制过程中需要的宿主蛋白,对发现新的药物靶标和研制新型抗流感病毒药物具有十分重要的意义。流感病毒的M2蛋白是一个多功能的蛋白分子,在病毒复制周期的多个环节发挥重要作用,目前鉴定的与M2蛋白相互作用的宿主蛋白仍为数甚少。本课题在前期工作中已经成功建立了筛选与M2蛋白相互作用宿主蛋白的酵母双杂交系统,拟进一步结合运用免疫共沉淀及质谱鉴定的方法全面筛选与M2蛋白相互作用的宿主蛋白。在此基础上,结合相关的功能研究确定宿主蛋白对流感病毒复制发挥的作用,阐明宿主蛋白在病毒复制周期中发挥作用的关键环节,揭示宿主蛋白对流感病毒感染性和致病性等生物学特性的影响。本项目的实施将深化对宿主蛋白参与流感病毒复制周期调控分子机制的科学认知,同时也为流感病毒的药物治疗提供新思路和新靶点。

英文摘要: Influenza virus is an important zoonotic pathogen. The current application of the anti-influenza drugs in use tends to result in the generation of drug-resistant strains. Thus, it has great significance to study the host proteins required for influenza virus replication for the discovery of new drug targets and the development of new anti-influenza drugs. Influenza virus M2 protein is a multifunctional protein molecule that plays an important role in many aspects of the viral replication cycle. Currently, very few host proteins have been identified to interact with the M2 protein. In previous work in this project, we have successfully established a yeast two hybrid system for the screening of host proteins interacting with the M2 protein, and will further combine the co-immunoprecipitation and mass spectrometry approach to comprehensively screen the host proteins that interact with the M2 protein. On this basis, we will employ the associated functional studies to determine the role of host proteins on influenza virus replication, to elucidate the key steps that host proteins exert their roles in the viral replication cycle, and to reveal the impact of host proteins on influenza virus biological characteristics such as infectivity and pathogenicity. The implementation of this project will deepen the scientific understanding on the molecular mechanisms of host protein functions in the regulation of influenza virus replication cycle, and will provide new thoughts and new targets for the treatment of influenza.
