中文摘要: 开发高效、快速、安全、经济的止血材料是临床手术、急救中急需解决的关键难点。本项目在前期研究发现低分子量蚕丝丝素多肽具有良好止血性能的基础上,拟从蚕丝丝素蛋白的可控水解着手,研究不同制备技术对蚕丝丝素多肽的成分与结构的影响,探索蚕丝丝素多肽分子量和结构的调控方法,构建分子量分布、结构、成分和止血性能四者之间的关系,阐明蚕丝丝素多肽的止血机制,研制适用于不同创面的蚕丝丝素多肽快速止血材料。本项目不仅为新型快速止血材料研究提供了新思路,而且还能通过对蚕丝丝素多肽结构的深入研究,加深对多肽生物材料的结构与功能之间的关系的理解,实现多肽结构的有效调控,为制备具有实际应用价值的蚕丝丝素多肽止血材料奠定理论基础,为解决现有的止血材料存在止血速度慢、副作用较大、生物相容性差、价格昂贵等问题做出贡献。

英文摘要: The development of an efficient, rapid acting, safe, cheap hemostatic biomaterial is identified as the most important unmet need in clinical surgery and first aid. In this study, the fibroin peptide are prepared through controlled hydrolysis of silk fibroin and the effects of different preparation techniques on the composition and structure of fibroin peptide are investigated based on the fact that the low molecular weight fibroin peptide derived from silkworm Bombyx mori has excellent hemostatic property in our previous study. The control method of the molecular weight and structure of fibroin peptide and the relationship among the molecular weight, structure, composition and hemostatic property are investigated. Furthermore, the hemostatic mechanism of fibroin peptide and the development of the fibroin peptide-based hemostatic biomaterials for different wound contours are studied. Our research will not only provide a new rapid acting hemostatic biomaterial, but also clarify the relationship between the structure and property of peptide-based biomaterial. We will control the structure of fibroin peptide effectively and establish the theory for the preparation of the fibroin peptide-based biomaterial for practical application.
