中文摘要: 皮肤组织作为机体与周围环境间的重要免疫屏障,蕴含了大量的免疫细胞。其中,常驻在皮肤组织内的DC,由于其功能的多样性,扮演了调节皮肤免疫应答的重要角色。皮肤中DC功能失衡与许多免疫性皮肤炎症的发病存在诸多联系。然而,DC在组织中的免疫调节功能及其诱导免疫耐受的机理尚不明确。银屑病是一种常见的、高复发性的慢性皮肤炎症。本项目拟以银屑病为模式疾病,在课题组所拥有的CD18减效突变小鼠银屑病模型基础上,建构由DC和CD4+ T细胞介导,发病率稳定的小鼠银屑病模型,藉以探索皮肤DC在炎症皮肤组织中的免疫调节功能,并揭示DC在诱导免疫耐受中的潜在分子机理。本项目也将深入探讨CD18这一重要的细胞黏附分子在DC诱导免疫耐受过程中的潜在功能。明晰组织中DC的免疫调节机制,将对现有DC生物学带进突破性的新观点,并成为未来诊治银屑病乃至其它自身免疫性疾病的重要基础。

英文摘要: The skin, as the primary interface between the body and external environment, harbors a high frequency of immune cells, among which DCs, because of their functional plasticity, play a critical role in the regulation of skin immune responses. Dysregulation of skin DCs is implicated in the pathogenesis of inflammatory skin diseases. Thus far, the immune regulatory property of tissue resident DCs and their capacity to mediate tissue immune tolerance remain unclear. Psoriasis is a common, relapsing chronic inflammatory skin disease. Taking advantage of our CD18hypo mouse model of psoriasis, through establishing a new DC and CD4+ T cell mediated psoriasis mouse model, this project aims to investigate the immunoregulatory function of skin DCs under inflammatory conditions, and to elucidate the molecular mechanism involved in DC mediated immune tolerance induction. Understanding the immunoregulatory mechanism of tissue derived DCs in target tissues may bring novel insight into DC biology and provide the base for future diagnosis and immune interventions in psoriasis, possibly aslo in other autoimmune inflammatory disorders.
