中文摘要: 前期研究发现,剧烈运动特别是离心运动导致骨骼肌收缩能力下降和超微结构改变,同时可诱导骨骼肌微管解聚,并伴有线粒体的结构和功能改变。针刺可减轻运动所致的微管和线粒体改变。运动导致的骨骼肌线粒体分布、形态及功能的改变必将对骨骼肌细胞凋亡和自噬产生重要影响,因此探究它们在运动性骨骼肌损伤中的作用具有非常重要的意义。Omi/HtrA2是作用较强的线粒体促凋亡蛋白,同时参与自噬的调节,但其在骨骼肌中的研究未见报道。 我们发现,离心运动可以导致骨骼肌Omi/HtrA2蛋白表达增加,且针刺能使Omi/HtrA2蛋白表达明显降低,提示Omi/HtrA2可能通过诱导细胞凋亡和自噬来加重骨骼肌损伤。本项目拟以Omi/HtrA2为切入点,探究运动引起骨骼肌细胞凋亡和自噬的变化机制,从细胞凋亡和自噬的角度探究运动性骨骼肌损伤的机理。同时探寻针刺治疗运动性骨骼肌损伤的效果及其可能机制。

英文摘要: Previous study has found that strenuous exercise is characterized by eccentric exercise led to the decrease of the skeletal muscle contraction ability and ultrastructure changes, at the same time can induce the microtubule depolymerization of muscle, and accompanied by the structure and function of mitochondria changed. Acupuncture can relieve the change of microtubules and mitochondrial caused by exercise. It is can't be ignored the change of mitochondrial distribution, morphology and function of skeletal muscle caused by exercise. And mitochondrial changes will produce important influence on apoptosis and autophagy. So it is very important to explore the roles in exercise-induced skeletal muscle injury. Omi/HtrA2 is the protein in mitochondria which is used to promote apoptosis with strong function and to participate in the regulation of autophagy, but the study did not reported in skeletal muscle. We found that the Omi/HtrA2 protein is increased in skeletal muscle by eccentric exercise, and the expression of Omi/HtrA2 can significantly reduce by acupuncture. Omi/HtrA2 may increase skeletal muscle injury through induction of apoptosis and autophagy. This project intends to use the Omi/HtrA2 as the breakthrough point to explore the changed mechanism of apoptosis and autophagy of skeletal muscle movement, and to explore the mechanism of exercise-induced skeletal muscle damage from the perspective of apoptosis and autophagy. At the same time, explore the effect of acupuncture in the treatment of exercise-induced skeletal muscle damage and its possible mechanism.
