中文摘要: 液晶分子拥有独特的液-固相性质,其长程有序性和光学各向异性能将化学和生物结合事件转换成放大的光学信号。基于液晶分子特性而发展起来的液晶生物传感器,具有构造简单、易于实现微型化、阵列化和可视化等特点,是一类极具潜力的新兴传感技术。近年研究发现,双链DNA分子的错配碱基对能特异性识别重金属离子Hg2+(T-Hg2+-T)和Ag+(C-Ag+-C),国际上有关此类分析检测重金属离子的研究近几年来也获得了飞速的发展。本项目拟结合错配碱基对特异性识别重金属离子的原理,设计出一系列DNA识别探针,研究新型液晶生物传感技术,实现同时高选择性分析检测环境中多种重金属离子;并进一步结合纳米粒子标记核酸分子,增强扰乱液晶分子取向排列效应,实现重金属离子的高灵敏分析检测。通过本项目的研究,将建立一种高选择、高灵敏、高通量的液晶生物传感分析技术,实现对环境中重金属离子以及其它小分子的分析检测,并探索其初步应用。

英文摘要: Liquid crystals (LCs) possess unique liquid-crystalline phase properties, and the long-range order inherent and optical anisotropy can transform chemical and biomolecular binding events into amplified optical signals.The liquid-crystal based biosensor has some features, such as simple structure,be easy to miniaturization and array.Recent studies have shown that the double-stranded DNA mismatch base pairs can specifically recognize the heavy metal ions.For examples, the thymine-Hg2+-thymine (T-Hg2+-T) interaction between the mismatched T-T base pairs and the cytosine-Ag+-cytosine(C-Ag+-C)interaction between the mismatched C-C base pairs.Recently,such analysis and detection of heavy metal ions has been developed rapidly in the international field. This project intends to combine the principle of specific recognition of heavy metal ions by the mismatch base pairs with the property of LC biosensor. We try to design a series of DNA recognition probe and achieve the aspiration of highly selective analysis and detection of heavy metal ions at the same time. It can also been achieved the purpose of highly sensitive analysis and detection of heavy metal ions by nanoparticles marked on the nucleic acid molecules, which can greatly disrupt the orientational arrangement of LCs.
