中文摘要: 小型猪在解剖学、生理学和疾病发生机理等方面与人在比较医学上极其相似,是理想的实验动物模型和异种器官移植供体。小型猪麻醉和颉颃苏醒是兽医学、比较医学和生命科学等领域急待解决的问题。本课题组在前两项国家自然科学基金资助下成功研制出小型猪专用复合麻醉及其颉颃剂,但对其机理研究甚少,因此需进一步研究其作用机制。近年来研究表明细胞内信号转导系统与全麻及催醒机制有密切关系。为此本项目拟进一步从信号转导系统MAPK、mTOR和Orexins系统入手,选取MAPK和mTOR信号转导系统及Orexins系统中关键蛋白及其下游转录因子为研究对象,研究注射小型猪专用复合麻醉及其颉颃剂后三个系统发挥的作用途径,确定小型猪专用复合麻醉及其颉颃剂的交互作用机制,并且观察阻断这些信号转导系统后,小型猪专用复合麻醉及其颉颃剂对小型猪麻醉和苏醒过程中临床指标安全性的影响,为兽医学、比较医学和生命科学的研究奠定坚实的理论。

英文摘要: Miniature pig was ideal experiment animals model xenotransplantation donor because of its similar with anatomy, physiology and mechanism of diseases in comparative medicine. The anaesthesia and antagonism of miniature pigs were the urgent needed to resolve problem in veterinary medicine, comparative medicine and life science. Our group had developed combined anaesthesia for miniature pigs and specificity antagonist for miniature pigs' combined anaesthetic, which were supported by two National Natural Science Foundation of China, but the mechanism of these were not clarification, so the mechanism of these drugs were needed to further research. Intracellular signal transaction had affinity relationship with mechanisms of general anaesthesia and antagonism. This program will research MARK, mTOR and Orexins signal transaction system, their key proteins and lower transcription factors were studied. Pathways of MARK, mTOR and Orexins signal transaction system were researched after combined anaesthetic for miniature pigs and specificity antagonist for miniature pigs' combined anaesthetic was administrator, in order to certify interaction mechanism of anaesthetic and antagonist.
