中文摘要: 氧化应激是癌症、心脑血管疾病和老年性痴呆等多种疾病的主要原因,评价和筛选具有强抗氧化活性的天然资源是目前急待解决的问题。红枣红色素是一种含有黄酮类化合物的水溶性色素,本项目拟以红枣红色素为研究对象,通过化学模拟试验和H2O2诱导氧化应激细胞损伤模型研究红枣红色素的体外抗氧化活性,建立小鼠常压缺氧、KBrO3诱导氧化应激性肾损伤、拘束应激诱导氧化应激性肝损伤动物实验模型,研究红枣红色素对氧化应激发生时机体内抗氧化酶和抗氧化物的改变,线粒体功能、线粒体膜电位变化的影响及其剂量-效应关系,探讨红枣红色素改善氧化应激的可能作用机制,以期从整体动物水平评价红枣红色素的抗氧化应激作用,阐述其抗氧化应激作用机制,以明确其生物活性,为红枣资源的综合利用、天然红色素的开发应用、抗氧化剂的筛选提供基础研究和理论依据。

英文摘要: Oxidative-stress is the main reason of causing cancer, Cardiovascular System disease, Alzheimer's disease, etc. To select and evaluate antioxidatant active natural resource is an urgent problem need to be solved at present. The red pigment of jujube is a kind of water soluble pigment containing flavonoids. In this project, we take red pigment of jujube as the research object, study antioxidatant-activity of the red pigment in vitro based on the chemical simulation and H2O2 induced cellular oxidative-stress injury model, and establish mice normobaric hypoxia, KBrO3 induced kidney injury and restraint stress induced liver injury animal test models respectively, and then analyze the effect of red pigment on antioxidative enzymes, antioxidant, mitochondrion function, membrane potential's variety, and dose effect while oxidative-stress injury occurred. All these will help to discuss the possible mechanism of the red pigment improving oxidative-stress status. We expect to evaluate Oxidative-stress effect of the red pigment in the whole animal level and elaborate the mechanism of antioxidative-stress from the perspective of biochemistry that will provide the basic study and theoretical basis for the comprehensive utilization of jujube, and the development of natural red pigment and the screening of antioxidant.
