中文摘要: 在纳米材料生物安全性研究中,最基础科学问题之一是理解纳米材料如何参与细胞代谢过程并且与代谢关键功能性生物大分子相互作用。在发展高通量微流控芯片并应用于生化检测的前期工作基础上,本项目集成高通量微流控芯片样品前处理技术与功能纳米材料,并结合质谱分析,建立发展在单细胞水平上研究纳米材料代谢过程与检测的方法学。设计微流控芯片实现大规模并行的单细胞隔离,利用纳米颗粒进入单细胞内原位地捕获富集代谢关键功能性生物大分子。运用磁性分离技术,将由纳米颗粒富集的生物大分子在芯片辅助下转移至基质表面原位地形成待检物的点阵列,应用MALDI TOF质谱进行高灵敏、高通量的检测。本项目发展的技术作为一种新的工具,不仅可以对纳米材料在单细胞的代谢过程进行研究,还可以应用于血清样品中的快速、多指标检测和临床疾病诊断。

英文摘要: It is a fundamental key question in examing biosafety issues of nanomaterials to understand the metabolic behaviors of nanomaterials, and the interactions between nanomaterials and the functional proteins and peptides involved with cellular metablism. Combined with mass spectrometry, this proposal integrates high-throughput microfluidic technologies for sample pretreatment and functional nanomaterials to develop the methodology of highly sensitive detection of biomacromolecules at the single cell level. The new design of microfluidic devices will allow parallel,large scale isolation of single cells;nano particles will be employed to capture and enrich the key functional proteins and peptides in cellular metabolism in situ. Magnetic separation will be applied in order to transfer these functional biomacromolecules to the substrate, forming arrayed analytes for MALDI-TOF mass spectrometry. As an important new tool, the technology by this proposal will facilitate the research on the metabolism of nanomaterials in single cells, but also can be applied onto rapid, multi-parameter dignosis of disease status using clinical serum samples.
