中文摘要: 在低氧训练备受重视的同时,很少有人关注低氧对运动性肌损伤(EIMD)的影响及其作用机制。我们前期研究发现,在低氧环境下运动或休息时,均会出现EIMD或类似的肌细胞损伤,表明除了机械应力牵拉之外,低氧刺激也可导致或加剧肌纤维膜完整性下降和膜骨架蛋白dystrophin的丢失,其作用机制引起我们极大的研究兴趣。本研究拟在前期研究基础上,通过建立离心运动诱导的EIMD动物损伤模型,采用EBD荧光法、免疫组化、Western blot、qRT-PCR、基因芯片和northern blot等检测方法以及信号通路抑制剂手段,分别从dystrophin的蛋白降解和合成调控、utrophin的代偿功能以及dysferlin、 calpain3的膜修复等多个途径,进行低氧对EIMD膜损伤影响机制的研究和探索,为低氧训练中运动性损伤的预防和合理控制、以及肌肉疾病的运动治疗提供理论依据。

英文摘要: Numerous studies have focused on the profit of hypoxic training. However, only a few of them concerned the hypoxic effect on exercise-induced muscle damage(EIMD)and its mechanisms. Our previous finding on the EIMD or similar damage induced by the exercise or rest under hypoxic environment showed that in addition to the mechanical stress, the hypoxic stress could induce or aggravate the changes of the sarcolemma integrity and dystrophin, a cytoskeletal protein. It is of interest to reveal the mechanisms of sarcolemma damage under hypoxia after exercise. In this study, we aimed to test the proposed mechanisms of sarcolemma damage effected by hypoxia through the degradation and synthesis of dystrophin, compensatory role of utrophin, membrane repair of dysferlin and calpain3,using EBD fluorescence, immunohistochemistry, western blot, QRT-PCR, gene chip, northern blot and inhibitors of signal pathway in EIMD rats model induced by eccentric exercise. The results would provide an important reference for prevention and control of EIMD in hypoxic training, and also the exercise therapy for muscle disease.
