中文摘要: 猪繁殖与呼吸综合症是世界流行性重大传染疾病,其根源为猪繁殖与呼吸综合症病毒,目前该病毒存在不同致病性毒株并很容易发生变异。本项目拟设计合成不同荧光发射波长的无镉水溶性量子点,采用带有活性基团的聚合物、链霉亲和素、配合物等对量子点表面进行化学修饰。利用不同致病性病毒毒株抗体与量子点进行化学偶联,筛选具有高效荧光猝灭能力的猝灭剂,猝灭量子点-抗体复合物荧光以构建猪繁殖与呼吸综合症病毒捕获探针,通过抗原抗体特异性相互作用后量子点荧光回升强度实现对不同致病性猪繁殖与呼吸综合症病毒毒株定性定量检测。并利用无镉量子点实现对致病性不同的猪繁殖与呼吸综合症病毒的高效、稳定标记,用荧光成像法观察不同致病性猪繁殖与呼吸综合症病毒毒株侵染宿主细胞后在胞内的分布情况,为该类疾病的早期诊断提供有效科学依据。

英文摘要: Porcine reproductive and respiratory syndrome is a world epidemic disease which is caused by porcine reproductive and respiratory. Nowadays, there are different virus strains and they can easily mutate. The project plan to design and synthesis water-soluble, cadmium free quantum dots with different fluorescence emission wavelengths, chemically modify the surface of the quantum dots by polymers with active groups, streptavidin or other complexes. Chemically coupled virus antibodies with quantum dots as-prepared. Select a quencher quenching quantum dot- antibody complex fluorescence with efficient fluorescence quenching ability and develop a rapid detection method for detecting different porcine reproductive and respiratory syndrome virus strains by fluorescence recovery intensity. In addition, label the virus with cadmium free quantum dot and investigate different porcine reproductive and respiratory syndrome virus strains distribution in the host cell by fluorescence imaging, providing an effective supporting for early diagnosis of this disease.
