中文摘要: 为了提高肺癌的治疗效果,需要实现肺癌的早期诊断。肿瘤标志物能够指示癌症发生发展,为癌症的早期诊断提供了可能。因此,构建针对肺癌肿瘤标志物的生物传感系统具有重要的临床应用价值。由于性质稳定、便于组装,四连体核酶作为信号元件已被广泛应用于生物传感系统中。但是,四连体核酶的分子识别能力尚未被开发。本项目拟设计一种具有长颈环的四连体序列并尝试利用这种序列组装高活性四连体核酶。此外,长颈环在分子识别方面的潜力得到系统的研究,以便实现双功能(信号输出和分子识别)在四连体核酶中的统一。基于以上研究,一种新的生物传感器构建策略被提出并被应用于构建多种肺癌肿瘤标志物生物传感器。

英文摘要: To have a better treatment effect of lung cancer, it is necessary to diagnose lung cancer as early as possible. Tumor marker can indicate the pathogenesis and development of a cancer, which provides the possibility of diagnosing a cancer at its early stage. Therefore, biosensors for tumor markers of lung cancer are of great value in clinical applications. Because of its chemical stability and ease of assembly, G-quadruplex-hemin DNAzyme has been widely employed in biosening systems as a signal element. However, the molecular recognition function of G-quadruplex-hemin DNAzyme has not yet been exploited. In this project, a G-quadruplex sequence with long loop is designed and attempted to assemble G-quadruplex-hemin DNAzyme that possesses high activity. Moreover, the potential of long loop in molecular recognition will be systematically studied aiming to achieve the integration of the dual functions (signal acquirement and molecular recognition) in G-quadruplex-hemin DNAzyme. Based on the above investigations, a novel strategy for biosensor construction is proposed and utilized in fabricating diverse tumor marker biosensors for lung cancer.
