中文摘要: 近年来蛋白质组学方法越来越多地被应用到植物应答非生物胁迫的研究中, 鉴定出许多新的抗逆蛋白质, 揭示了参与胁迫耐受的蛋白质翻译后调控机制, 增进了对于植物耐受非生物胁迫分子机理的认识,同时为植物抗逆性机制研究提供了新的研究思路。蒙古冰草(Agropyron mongolicum Keng),2n=14,是旱生-沙生草原种,常见于干草原和荒漠草原的典型沙质环境中,是国家二级珍稀濒危植物和急需保护的农作物野生近缘种。本项目将在前期研究基础上,从蛋白质组学出发,进一步揭示蒙古冰草的抗旱机制,拟采用双向电泳技术和质谱分析技术对蒙古冰草在干旱胁迫下表达的蛋白进行差异分析,获得与干旱胁迫密切相关的蛋白质组份,进行序列分析,通过基因克隆获取重要差异蛋白基因,并进行功能验证。该项研究结果将揭示蒙古冰草参与干旱胁迫耐受的蛋白质翻译后调控机制, 为这一优良饲草的保护及开发利用奠定科学基础。

英文摘要: In recent years proteomics method is more and more applied to the study of plant abiotic stress response , many new stress-tolerant protein were identified, the stress tolerance of protein gene post-translational regulation mechanism were revealed, and enhanceto understand for the abiotic stress tolerance for plant molecular of the mechanism , these work can provide a new idea for plant resistance mechanism research. Mongolian wheatgrass (Agropyron mongolicum Keng) is xerophilous- Psammophyte species, 2n=14. and is the national secondary rare and endangered plant, need to be protected urgently as wild relatives of wheat in Desert Grassland and Typical Grassland of China. This project will be based on the previous studies, further study on the drought-resistant mechanism of Mongolian wheatgrass with the two-dimensional electrophoresis and mass spectrum technology of proteomics to analysis on expression differentail protein under drought stress, and get protein components to analyzing on sequence characteristics. The full length sequence of expression differentail protein gene will be cloned and identified their function.
