中文摘要: 人体内氧化应激产生的自由基是导致人类许多疾病的重要因素,利用天然产物中抗氧化活性物质对抗体内过量自由基是医学领域的主要手段。从天然产物中筛选抗氧化活性物质的关键科学问题是建立科学的评价方法。现有体内外抗氧化活性评价方法存在各种局限性,在面对天然产物这一复杂体系时,缺乏灵敏、快速、准确的评价方法,是制约从天然产物中评价和筛选抗氧化活性物质的关键技术瓶颈。本研究拟采用细胞代谢组学的研究策略,以HPLC-DAD-MS为技术平台,结合化学计量学手段,构建基于细胞内源性代谢物为生物靶标,与目标化合物、生命体系高度关联的定量数学模型,发展快速、灵敏、高选择性的抗氧化活性物质评价新方法。在此基础上,以油橄榄叶为研究对象,评价和筛选其抗氧化活性物质,阐明其物质基础。通过本项目实施,为从天然产物中评价和筛选抗氧化活性物质提供技术支持,为阐明天然产物中抗氧化活性物质基础提供理论指导。

英文摘要: Free radicals produced from oxidative stress are the important factor that leads to various diseases and the antioxidants from natural sources are used to neutralize excess free radicals in the field of medicine. The key problem of screening antioxidants from natural sources is to establish the scientific evaluation method. However, there are many inherent defects, such as low sensitivity, poor accuracy and slow speed, in the existing methods for the evaluation of antioxidant activity, which hamper the research on screening antioxidants from natural sources. In this project, a quantitative mathematical model will be constructed based on cellular metabolomics and HPLC-DAD-MS methods, which describes the relationship between antioxidants and metabolite targets, and then a new method for the evaluation of antioxidant activity, with the properties of high sensitivity, high selectivity and rapid response time, will be established. This new method will be applied for the screening of antioxidant ingredients from olive leaves and the material basis of olive leaves will be elucidated. The implementation of this project will undoubtedly provide technical support for the screening of antioxidant ingredients from natural sources and provide theoretical guidance for elucidating the material basis of natural products for antioxidant effect.
