中文摘要: 沙漠小球藻是在单细胞水平上研究水盐胁迫条件下低等植物基因表达及其调控特征的重要材料之一。这方面的研究日渐增多。本研究利用从新疆沙漠中分离、纯化培养的小球藻,通过建立其水和盐胁迫模型,运用数字基因组表达谱分析(DGE)、实时定量PCR(qPCR)、Northern Blot、Western Blot、蛋白质二维电泳等技术方法,研究和分析比较在水盐胁迫条件下沙漠小球藻胞内糖和蛋白质等组成型成分含量及其变化规律,胞内信号感受与传递途径相关调控基因的诱导型表达及其调控特征,包括藻细胞转录组水平、胞内IAA/ABA含量、LEAs蛋白(Ccor1/Ccor2、Cw80、Lea3),sHSPs以及相关转录因子(BhHsf1、CpHB-7、BhWRKY1)实时表达变化规律。为单细胞低等植物向复杂高等植物进化过程中,在新疆特定生境下,水盐胁迫应答分子机制及其演进提供实验证据。

英文摘要: Chlorella is a perfect material for studying gene expression and regulation characteristics of plant, which is under water and salt stress, at the single cell level. At present, the related researches in this area are growing. In this study, through establishing the model of water and salt stress using Chlorella from desert of Xinjiang, we will study and analyze the varied regular patterm of the desert Chlorella's intracellular glucose and protein content, the expression and their regulating characteristics of genes related to intracellular signal's perception and the transmission pathways including the variation of transcriptome expressional level, the content of ABA hormone and LEAs protein and the varied regular patterm of the related transcription factors'(BhHsf1、CpHB-7、BhWRKY1)real-time expression by using the methods of Digital genome expression spectrum's analysis (DGE), quantitative real-time PCR, Northern Blot , Western Blot and protein's two-dimensional electrophoresis. The researches can provide experimental evidence for the molecular mechanisms of stress resistance and the processes of evolution from single cell lower plants to complex higher plants.
