中文摘要: 微生物油脂在功能食品、高级化工和医药等领域有替代动植物油脂的潜力。但产油微生物只在缺氮的培养条件下才能合成高水平的油脂,这限制了油脂的产量。目前尚不清楚氮饥饿通过何种分子机制实现与油脂合成的偶联。在本项目中,将敲除ACA-DC 50109菌株中氮阻遏转录激活蛋白YLGat1的编码基因,证实YLGat1在介导氮饥饿与油脂合成偶联中的作用;利用转录组学分析明确YLGat1介导氮饥饿与油脂合成偶联的分子机制;并通过超表达无磷酸化氨基酸位点的YLGAT1基因,构建解除氮饥饿与油脂合成偶联的油脂生产菌株。本项目的研究能进一步完善微生物油脂合成的分子机理,同时也为提高微生物油脂的产量提供了新思路。

英文摘要: Microbial oil has huge potential in fields of functional foods, advanced chemicals and medicines. But high-level lipids were only obtained in the nitrogen-staving conditions, which limits the lipid production. It was not clear in which molecular mechanism nitrogen starvation was coupled with lipogenesis. In this project, the gene coding for YLGat1 in Yarrowia lipolytica ACA-DC 50109 will be disrupted, to verify that YLGat1 plays a role in mediating the coupling between nitrogen starvation and lipogenesis. Then transcriptomics methods will be used to clarify the mediating mechanisms of YLGat1. Also the coding gene for YLGat1 without potential phosphorylated sites, will be overexpressed to construct a lipid producing strain ,in which nitrogen starvation was not coupled with lipogenesis. Sduty in this project can give more knowledge about the synthesis mechanism of microbial oil, and provide a new perspective to enhance lipid production.
