中文摘要: 在水相中分别合成具有超顺磁性的四氧化三铁磁性纳米粒子和具有荧光的金团簇,然后在微乳液体系中用生物相溶性好的二氧化硅包覆两种粒子,制备出同时具有超顺磁性、荧光和模拟酶特性的多功能材料。用偶联试剂在该材料表面偶联酶分子(如葡萄糖氧化酶、胰蛋白酶),在外磁场协助作用下,该材料可对溶液中相应酶底物分子(如葡萄糖、牛血清白蛋白)有效地分离富集。富集在材料表面的酶和底物反应产生的过氧化氢可淬灭复合材料的荧光,从而可根据荧光信号的变化实现对目标分析物的荧光可视化检测;同时,四氧化三铁和金团簇均具有模拟酶特性,在反应产生的过氧化氢协同作用下,能够酶催化过氧化酶底物产生有色物质,因而可用比色法直接定量分析过氧化氢,达到间接定量分析目标分析物目的。该项目实现了在同一载体上对目标分析物的有效分离、富集、可视化定性荧光分析及比色定量分析,有效提高了分析物的灵敏度,在野外现场分析及临床分析中具有好的应用前景。

英文摘要: Superparamagnetic nano Fe3O4 and fluorescent Au clusters were prepared firstly in the aqueous phase. Next, two particls were encapsulated with good biocompatibility SiO2 in a microemulsion system. The prepared multifunctional material is superparamagnetism, fluorescence and mimic peroxidase. Enzymes, e.g. glucose oxidase and trypsin, can be coupled on the surface of the material, and the enzyme substrats, e.g. glucose and bovine serum albumin, can be seprated and enriched in the presence of external magnetic field. The fluorescence of material will be quenched by peroxid produced from enzymes and enzyme substrats, and the fluorescent signal are used for fluorescent detection for targent substract. Meanwhile, Fe3O4 and Au clusters exhibited intrinsic peroxidase-like activity, which catalyze the oxidation of a peroxidase substrate by H2O2 to the oxidized colored product and provides a colorimetric detection of targent substract. The target analyts can be separated and enriched and visual qualitative analysis and quantitative colorimetric analysis on the same vector in this project. This method is potentially useful in visual detection of biomolecules in field analytical chemistry and clinical analysis.
