中文摘要: 在对金纳米棒光学性质的理论和实验研究的基础上,创新性地提出一种在磁性纳米颗粒辅助下生物编码的金纳米棒探针多通道的生物传感和检测机制,并将该机制应用于急性冠状动脉综合征(ACS)生物标志物的特异性和多通道的检测。检测的实现是基于不同生物编码的多种金纳米棒在同一液相体系中的光学吸收峰仍彼此独立,并且每种峰强度与其数量成正比。修饰在金纳米棒上的特异性单克隆抗体仅与特定的标志物结合,在外加磁场作用下通过免疫磁性纳米颗粒的辅助将与标志物分子结合的金纳米棒快速分离。通过测量磁分离后的检测体系与参照体系的金纳米棒的吸收峰强度的变化反应标志物的种类和含量。上述水基的检测体系不需要繁琐费时的分离和洗涤过程,因而优于生物检测中的一些传统方法。本课题的研究还兼具普适性和与临床诊断相结合的特点,可以进一步拓展为诊断同一疾病的不同阶段或同步诊断不同的疾病,极大方便临床应用。

英文摘要: Based on the theoretical and experimental results of the optical characters of gold nanorods, we propose a novel strategy for multiplex detection of acute coronary syndrome (ACS) biomarkers using biocoded gold nanorod probes by the assistant of magnetic nanoparticles. This strategy lies in the longitudinal surface plasmon resonance (SPR) peaks of biocoded gold nanorods with proper aspect ratios can keep independent in a same solution and the intensity of each longitudinal peak is proportional to the quantity of the corresponding gold nanorods. With the help of immunomagnetic nanoparticles under external magnetic field, gold nanorod-antibody-biomarker complexes would be quickly separated and the presence as well as content of biomarkers can be detected by comparing the intensisty changes of the longitudinal SPR peaks before and after magnetic separation. This strategy is no need of time-cost washing or spearating circles such as conventional ELISA method and very suitable for clinic applications.
