中文摘要: 鲟鱼属软骨硬鳞鱼类,在电感受器的进化中占据着极为重要的地位。本项目在前期对西伯利亚鲟侧线系统及早期发育研究基础上,首次利用细胞标记Dil,追踪西伯利亚鲟早期胚胎期侧线基板细胞的分化和转移,研究侧线感受器的形成和其伴随着的神经末梢的进入;项目通过免疫组织化学与现代分子生物学(荧光定量PCR、RACE、Northern blotting和原位杂交)等技术进一步深入研究Sox2和Sox9基因功能、表达和定位,初步探明Sox2和Sox9基因对西伯利亚鲟早期侧线发生的调控机制;应用透射电镜技术研究西伯利亚鲟侧线发育不同时期侧线感受器的超微结构,从结构上阐明侧线发育机制。预期研究结果不仅可以直接促进鲟鱼类侧线发生发育调控机制的研究进展,而且对于认识电感受器起源与进化具有重要理论和实践意义。

英文摘要: The Siberian sturgeon (Acipenser baerii Brandt), a chondrostean, occupies an important position in the evolution of the electroreceptor. In our previous study, we had done researches on the lateral line system and early development of Siberian sturgeon. In this project, we will label the lateral line placode with Dil, to track the formation of the lateral line receptors and the associated entry of nerve endings. We further study Sox2 and Sox9 function, expression and localization using Immunohistochemical and modern molecular biology techniques (fluorescence quantitative PCR, RACE, Northern blotting and in situ hybridization), with the purpose of ascertaining their regulatory mechanisms toward early development of the lateral line. The development of lateral line system during early ontogeny of the Siberian sturgeon will been examined by using Transmission Electron Microscopes (TEM), Ultrastructure of the lateral line should elucidate the mechanism of the development of lateral line system. The expected results can not only directly contribute to the progress of studies on the regulation of the lateral line development in sturgeons, but also have important theoretical and practical significance for understanding the origin and evolution of electroreceptor.
