中文摘要: 研究表明MSTN对肌内肌卫星细胞和前脂肪细胞的脂肪分化存在差异,但差异的原因并不清楚。我们推测在肌卫星细胞中MSTN通过诱导MyoD的表达,进而抑制PPARγ的转录;而在前脂肪细胞中,MSTN并不能诱导MyoD抑制PPARγ的转录,进而导致MSTN对两种细胞的脂肪分化存在差异。因此本研究中首先通过Realtime-PCR、Western blot、细胞免疫化学染色等确定两种细胞中MyoD和PPARγ表达是否存在差异;接着利用生物信息学预测、EMSA、Chip等试验确定MSTN在肌卫星细胞中是否通过上调MyoD的表达抑制PPARγ的活性,而在前脂肪细胞中MSTN并不能上调MyoD的表达抑制PPARγ转录。从而导致MSTN处理后的肌卫星细胞和前脂肪细胞脂肪分化存在差异。本项目若被证实将揭开MSTN对不同细胞类型脂肪分化差异的原因,同时还可揭示MSTN对肌内脂肪的形成影响和与之相关疾病形成原因。

英文摘要: Researches indicated that MSTN had different regulation roles on the adipogenesis of Muscle satellite cells and Preadipocytes, but the reason is not clear. We conjecture that MSTN could induce the expression of MyoD to inhibit the transcription of PPARγ in Muscle satellite cells, which is incorrect in Preadipocytes. In this research, we explore whether the expressions of MyoD and PPARγ are different in Muscle satellite cells and Preadipocytes after treated by MSTN using Realtime-PCR、Western blot and Immunocytochemistry. Then we determine whether PPARγ could be inhibited by MyoD using bioinformatics, EMSA, Chip in Muscle satellite cells but not in Preadipocytes. Thus MSTN could have different functions on the adipogenesis of the two types of cells. If this hypothesis is confirmed, it will tell us why MSTN had different functions on the adipogenesis of different type of cells. The source of adiposities in the muscle and the related disease also will be resolved.
