中文摘要: 阿尔茨海默病(Alzheimer's disease,AD),是一种中枢神经系统退行性疾病,发病机制尚未明确。近年研究发现,血管因素在AD发生中占重要作用,认为脑血管损伤引起血脑屏障(BBB)功能失调,进而引起Aβ等神经毒性分子在脑内聚积,最终导致神经元损伤和神经系统退行性改变。有报道指出Slit2 过表达小鼠具有脑血管通透性改变及脑内Aβ表达增多的双重表现,我们的前期研究亦发现Slit2 过表达小鼠具有类似AD的行为学改变。因此,本项目将以Slit2-Tg小鼠,Tg2576小鼠及两者杂交产生的双转基因小鼠为基础,以脑血管内皮细胞通透性改变为切入点,研究Slit2转基因小鼠出现类似AD的症状,与小鼠BBB的结构和功能的改变,体内Aβ转运的变化和沉积之间的关系,阐明Slit2过表达小鼠导致AD的原因,为AD研究提供一种新型的模型小鼠。

英文摘要: Alzheimer's disease (AD) as a central nervous system degenerative disease,which pathogenesis is notclear. Recent studies have shown that vascular factors play an important role in AD development and formation that cerebrovascular damage caused by blood-brain barrier (BBB) dysfunction,causing neurotoxic Aβ molecules accumulate in the brainand resulting in nerve neuronal injury and neurological degenerative changes. Overexpressing Slit2 mice has cerebral vascular permeability changes in the brain and increasing Aβ expression,So far our study also found that over-expression of Slit2 mice have similar behavioral change AD like. Therefore, this project will use Slit2 mice, Tg2576 mice and their hybrids focusing on cerebral vascular endothelial cell permeability changes, to study Slit2 transgenic mice AD-like symptoms and changes of the BBB permeability, and the relationship between the Aβ deposition and BBB. We maybe clarify the reason Slit2 over-expression in mice leads to AD like and provide a novel mouse model for AD study.
