中文摘要: 探索简单、快速,能够实现复杂样品中多组分分析的传感方法是当今分析科学研究的热点和挑战。本项目从病原微生物检测在公共卫生安全保障等方面的迫切需要出发,拟设计一系列基于核酸构象变化及各种核酸工具酶作用的等温信号放大技术,筛选具有合适形貌、尺寸以及良好稳定性和生物相容性的纳米材料作为SERS基底或探针载体材料,同时优化寡核苷酸固定化方法。通过表面增强拉曼散射光谱检测手段,构建超灵敏、高特异性、稳定性好的分析方法,实现对大肠杆菌、沙门氏菌、丙型肝炎病毒等常见病原微生物的同时快速检测。本项目将充分利用核酸等温信号放大技术与表面增强拉曼技术的优势,进行测定方法上的改进和创新。同时,为生命科学领域中大量生化物质的检测提供有价值的参考和技术支持。

英文摘要: The development of biosensors for simple,direct and multiplex detection in the complex samples is the challenge of today's analytical scientific research. Present project focuses on the urgent need of pathogen detection in public health security, intends to design several isothermal amplification technologies based on nucleic acid conformation change and various tool enzymes, choose the perfect nanomaterials which have suitable morphology, size, good stability and biocompatibility used as the SERS substrates or probes, and optimize the immobilized methods of oligonucleotides. Combining with surface-enhanced raman spectroscopy, this project intends to construct some detection methods for rapid and multiplex detection of pathogens such as Escherichia coli, salmonella and hepatitis c virus with super sensitivity, high specificity and good stability. This project will take full advantages of nucleic acid isothermal amplification technologies and surface-enhanced raman spectroscopy to improve and innovate the detection methods. Meanwhile, the project will provide a valuable theory and technical support for the biochemical detection in the field of life science.
