中文摘要: 药用植物皂角刺具有多种生物药理活性,近年来受到国内外学者的广泛重视。作为我国中医治疗癌症的常用药之一,通常认为皂角刺含有的三萜皂苷和黄酮是该植物药抗癌作用的物质基础。本课题组前期研究,从皂荚刺的抗癌活性部位中分离得到一类黄烷酮骨架化合物,体外具有显著的选择性的抑制癌细胞生长作用,对这一传统中药的科学基础提出了新的研究方向。项目拟在前期研究的基础上,对药用植物皂荚刺中的黄烷酮骨架类化合物进行系统的分离纯化,通过体内外抗肿瘤活性实验,筛选出具有低毒高效的抗癌黄烷酮骨架的新的化合物衍生物;同时,阐明黄烷酮骨架结构的抗肿瘤作用机制和构效关系,揭示皂角刺抗癌机理,进一步为植物药靶向分子的研究提供科学依据。

英文摘要: As a medicinal plant with variety biological and pharmacological activities, Spina Gleditsiae attracts great interests of both of national and international scholars in recent years. It is also one of the commonly used drugs for cancer in traditional Chinese medicine. The flavone and saponin components were previously known as the material basis for the anticancer activity of Spina Gleditsiae. Beyond the current understanding, we have isolated several flavanone skeleton compounds with selective anticancer effects from Spina Gleditsiae in the preliminary study. Based on this, the proposed study will further systematically isolate and identify the flavanone constituents of Spina Gleditsiae. The novel flavanone skeleton compounds with selective anticancer effects will be screened in both of in vitro and in vivo models. Their anticancer effects and structure-activity relationship will also be investigated to elucidate the anticancer mechanisms of Spina Gleditsiae, thus providing the scientific bases for the further research of the herbal targeted molecules.
