中文摘要: 癌症是当今严重威胁人类健康的主要疾病之一。早期诊断并及时治疗是预防、治愈恶性肿瘤的关键。纳米材料的蓬勃发展为肿瘤的早期诊断提供了新的机遇。其中荧光贵金属纳米簇具有超小的尺寸、良好的生物相容性和稳定的发光性质,已成为生物、医学领域最理想的活体荧光标记物之一。然而,这方面的研究仍处于起步阶段,在贵金属纳米簇的功能化方法、多靶标功能化和多功能监测和生物成像等方面还亟需进一步研究。本项目拟以重要肿瘤标志物和肿瘤细胞成像为目标,从简化荧光金纳米簇的功能化方法、多靶标功能化双色荧光金纳米簇探针和集癌细胞成像与药物释放和治疗为一体的多功能金纳米簇荧光探针的构建及在癌细胞成像中的应用等方面进行研究。本项目的实施可望拓展贵金属纳米簇在生物成像领域的应用,丰富构建多功能贵金属纳米簇探针的策略,实现对肿瘤标志物的准确、灵敏和多目标同时监测,还可为构建肿瘤成像与治疗一体化的应用模型提供新思路。

英文摘要: Cancer is nowadays a serious threat to human health. Early diagnose and timely treatment is the key to prevent and cure the malignant tumor. Nanomaterials provide a new way for tumor imaging. Fluorescent noble metal nanoclusters (NMNC) offer many attractive features as ideal fluorescent labels for biological and medical applications, such as ultra-small size, good biocompatibility and excellent photo-stability.However,further development of simple ways for fabricating functionalized NMNC, multifunction of NMNC for simultaneous detection and imaging of multitargets and integrating diagnosis and therapy is highly needed. This project aims at developing a one-pot approch for fabricating functionalized gold nanoclusters (AuNCs) for cancer cell imaging, multifunctional AuNCs probe for targeting and imaging of multi cancer biomarkers, and multifunctional AuNCs probe for both diagnosis and therapy of cancers. The project will broaden the applications of NMNC in bioimaging, add novel strategies for designing multifunctionalized NMNC probes, and provide new methodologies to assemble imaging and therapy in one NMNC probe.
