中文摘要: 主要组织相容性复合物(MHC)具有高度多态性,基于表位肽的基序特征定义MHC 分子超型,是研究肽与MHC分子的相互作用机理以及基于抗原表位设计疫苗进行疾病防治的理论基础。目前MHC I 分子超型定义相对完善,而MHC II 分子超型定义不明确。可以和多个MHC 分子结合的表位即泛宿主表位,与一般表位相比包含了更多的生物信息,本研究拟筛选现有表位数据库中的泛宿主表位,建立一个泛宿主病毒表位数据库;根据泛宿主表位与MHC 分子的结合特征按照存同求异的原则分类,对相应的差异序列进行比对分析,研究泛宿主性差异产生的序列基础。对MHC II 类分子相关泛宿主表位分析寻找更准确定义MHC II 分子超型的新方法;对I 类分子相关泛宿主表位(简并表位)分析研究表位肽MHC 非严格限制性的序列特征,并采用分子动力学实验模拟计算,为表位改造以及预测疾病的易感性提供理论基础。

英文摘要: The development of the epitope-based vaccine, which have wide population coverage, is greatly complicated by MHC polymorphism. The grouping of the alleles into supertypes, on the basis of common structural and function features, address this problem directly. Furthermore the definition of the MHC supertype gives us a more unambiguous comprehension about the mechanism of the reaction between the MHC molecules and the binding peptides. Now the clusters of class I HLA molecules have been well defined, while the class II HLA molecules have not a well result. Promiscuous epitope which could be bound to more than one allele, may contain more information than general epitope. In this work, we will collection promiscuous epitopes from the available epitope database on the internet web, and build a promiscuous epitope database. Sequence analysis and alignment of these epitopes on the basis of the MHC-peptide complex structure feature may provide some suggestion on the definition of class II HLA molecules and a clear comprehension about the sequence basis of degenerate epitopes of class I HLA molecules. For special epitope , the molecular dynamic simulation of the corresponding pMHC complex will be done with amino acid mutation at special position.
