中文摘要: 很多研究表明,纳米药物载体的形貌和尺度对其给药性能具有重要影响。羟基磷灰石(HAp)是一种理想的药物载体材料,但是关于HAp的形貌对其作为纳米药物载体性能的影响规律尚不清楚。目前还没有一个关于HAp纳米药物载体的设计原则,到底哪种形貌能够获得最好的给药效果至今还是一个困扰学术界的疑问。基于纳米片状无机药物载体的独特优势,本项目拟在完成二维片状HAp形貌和尺度调控的基础上,以阿霉素(DOX)为模型药物,考察二维片状HAp及相近粒度条件下的球状、棒状HAp对药物的不同吸附和释放性能;以人的骨肉瘤细胞系MG63为模型细胞,研究不同形貌HAp复合药物载体被肿瘤细胞摄取的动态过程及对肿瘤细胞的抑制效果。该研究旨在揭示纳米HAp在药物负载、释放和细胞摄取方面的形貌学效应,可以在一定程度上对前人关于纳米粒子的尺寸和形貌效应理论进行补充和完善,对设计生物相容性好和高载药释药效率的新型载体具有重要意义。

英文摘要: Many studies have shown that morphology and size have important influences on the performance of nano drug carriers. Hydroxyapatite (HAp) is an ideal drug carrier material. However, it is unclear about the influnce of morphology and size on the performace of HAp drug carrier. So far, there is no design principle for HAp drug carrier, especially in morphology. Which morphology can achieve the best drug deliver effect is still a troubled academic question. On the basic of well control of size and morphology of lamellar HAp, The project intends to use doxorubicin (DOX) as a drug model and make an investigation on the drug adsorption and release properties of spherical, rod-like and lamellar HAp particle with similar size. In addition, this project intends to study the uptake dynamics process of HAp drug carrier by tumor cells and the inhibitory effect on tumor cells using human osteosarcoma cell line MG63 as a cell model. This study aimed to reveal the morphology effect of nano HAp on drug loading, drug release and cellular uptake. Also, in a certain extent, this study can supplement and perfect the previous theories on the morphology/size effect of nano drug carriers and it is significance for the design of biocompatible and efficiency drug carrier.
