中文摘要: 针对亲水性多糖类大分子和疏水性聚酯类高分子共聚合过程中存在的相容性差和非均相反应等问题,本课题以壳聚糖和聚乳酸为研究对象,试图借鉴非均相液体制剂制备的原理,提出采用乳化-自组装新方法构建一种两亲性壳聚糖/聚乳酸刷状共聚高分子。该方法首先将表面活性剂用于壳聚糖和聚乳酸混合乳剂的制备中,利用表面活性剂能显著降低液体之间表面张力的特性来解决两种溶液相容性差的问题;进而利用碳二亚胺活化羧基的方法,使混合乳剂中的壳聚糖和聚乳酸发生分子间自组装,以此来解决两种溶液之间的非均相反应。通过对共聚物进行结构分析、性能表征以及细胞相容性和组织相容性评价,揭示共聚物结构组成-理化性能-生物相容性之间的对应关系,并从理论上指导共聚物分子结构的优化设计,最终确立一套构建壳聚糖/聚乳酸两亲共聚物的新方法体系。

英文摘要: Poor compatibility and non-uniform phase reaction are major drawbacks that restrict the copolymerization between hydrophilic polysaccharide biomacromolecules and hydrophobic polyester. The purpose of present research project is to fabricate amphiphilic chitosan/poly(lactic acid) brush-like copolymer by using a novel emulsion and selfassembly technique. Firstly, in order to decrease the surface tension and improve the compatibility between two immiscible solution, surfactant is utilized in the preparation of chitosan and poly(lactic acid) based complex emulsion. Then, carbodiimide is used to active the carboxyl groups in complex solution and further induce the self-assembly reaction between the chitosan and poly(lactic acid). The reaction conditions are optimized to achieve the excellent copolymer composition. The obtained copolymer is characterized by structure analysis, properties tests and biocompatibility evaluation to explore the relationship between the structure composition and the biological properties. Finally, based on the results acquired in this research project, we will conclude a novel mechanism for fabricating the chitosan and poly(lactic acid) based amphiphilic copolymer.
