中文摘要: 10-羟基-2-癸烯酸(10-hydroxy-2-decenoic acid,10-HDA)是一种不饱和脂肪酸,是蜂王浆的主要活性成分,目前自然界中主要在蜂王浆中发现。研究表明10-HDA具有抗菌、免疫调节及抗肿瘤等多种重要的生理功能,应用前景广泛。10-HDA 主要分泌于蜜蜂工蜂的上颚腺,但其生物合成的分子机理目前还不清楚。本项目在分析工蜂上颚腺10-HDA分泌规律的基础上,应用比较蛋白质组学的方法对不同日龄工蜂上颚腺的差异蛋白进行分离和鉴定,筛选生物合成10-HDA的相关蛋白。同时对10-HDA生物合成途径中候选关键酶基因进行克隆与表达分析,并运用RNA干扰技术鉴定其在10-HDA合成代谢中的功能,明确调控10-HDA合成代谢的关键酶,揭示10-HDA生物合成的分子调控机制,为生物合成10-HDA奠定理论基础。

英文摘要: 10-hydroxy-2-decenoic acid(10-HDA),an unsaturated fatty acid,is the main active component of royal jelly, which has been mainly found in the royal jelly by now. It has been demonstrated that 10-HDA possesses remarkable biologic activities in antibacterial, immunoregulation, and antitumour. 10-HDA is mainly secreted from the mandibular glands of the worker honeybees, however, untill recently it is unclear about the underlying molecular mechanism of its biosynthesis. By analyzing the secretion pattern of 10-HDA, we would make separation and identification on the 10-HDA biosynthesis associated proteins from the mandibular glands of workers of different ages by comparative proteomics. Futhermore, the key enzymes in 10-HDA biosynthesis pathway would be identified by gene cloning, expression analysis and studying their functions using RNAi. These findings would demonstrate the molecular mechanism of 10-HDA biosynthesis and lay a theoretical foundation for 10-HDA biosynthesis.
