中文摘要: 血管生成与肿瘤等多种疾病的发生发展以及治疗密切相关。与基因组、蛋白组相比,血管生成过程引起的内源性代谢网络的变化为该类疾病的认识和治疗带来新的思路。斑马鱼模型用于研究血管生成作用与其他动物模型相比有明显优势。本课题用LC/Q-TOF与GC/MS检测技术建立斑马鱼代谢组学方法对药物干预斑马鱼血管生成过程前后进行对比研究,分析干预前后内源性代谢组及代谢轮廓的变化,通过数据的模式判别(PCA、PLS-DA)与统计分析,甄别与血管生成相关的代谢生物标志物,进一步通过代谢网络数据库(KEGG、HMBD等)以及生物信息学探索血管生成过程对机体代谢网络的关键节点的影响。本课题为研究血管生成疾病相关的重要生物标志物提供了新的研究思路,对恶性肿瘤等相关疾病的诊断和治疗提供借鉴。

英文摘要: Angiogenesis mediates numerous crucial pathologic and physiologic processes,such as cancer, inflammation, ischemia diseases and others. Comparision with genomics and proteomics, metabonomics related with angiogenesis proviede a new idea for clinic diagnosis and therapy of these diseases. Study angiogenesis effect using zebrafish has obvious advantage comparison with other animal models. LC/Q-TOF and GC/MS will be used to establish zebrafish-based metabonomics method, which will be applied to comparative research of natrual angiogenic process and its intervention with medications. Changes of metabonomics in angiogenesis course are indicated by mode determination (PCA and PLS-DA) and statistic analysis. Furthermore, some famous common metabonomics database combined with bioinformatics analysis contribute to indentify special metabolic biomarkers responsible for angiogenesis. The new method in this application will helpful to illuminate the modulation of metabolic network in organism by angiogenesis and important for diagnosis and therapy of angiogenesis related diseases.
