中文摘要: 癌症(恶性肿瘤)转移的预兆是具有侵袭性的癌细胞进入到外围血液或淋巴系统,因此从血液中分离出癌细胞,是阐明肿瘤转移机制与个性化诊疗的关键。本申请结合细胞生物学和电动力学,采用基于光诱导介电泳(ODEP)的微纳操控技术,首次提出将细胞旋转特性与受介电泳力的特性作为一类特殊的"生物标记",阐明细胞旋转和受力与非均匀电场梯度之间的关系。发展基于ODEP的癌细胞识别、筛选及分离技术,旨在实现一种面向淋巴癌早期诊断与个性化治疗的高通量、免标记无损细胞分离方法。具体包括:建立光诱导非均匀电场条件下的细胞极化和电液动力学模型,实现细胞精确、并行操控;研究肿瘤细胞与血红细胞免标记无损分离的机制和梯度电场的优化设计;开展ODEP芯片与聚合物PDMS微流管道集成方法研究,建立高分辨率的实验验证平台,实现B淋巴瘤细胞从正常B淋巴细胞和血红细胞中的筛选与分离。本研究对于推动我国癌症的临床诊疗水平具有特别重要的意义。

英文摘要: The concentration of cancer cells (malignant tumor cells) in the peripheral bloodstream or the lymphatic system is prognostic of metastatic disease in cancers. Thus, the separation of cancer cells from the blood is essential for clarify the mechanism of tumor metastasis and the individualized treatment. In this project, we study on the micro- and nanomanipulation with the combination of cell biology and electrokinetics based on optically-induced dielectrophoresis (ODEP). The cell rotation features and the difference of DEP force induced by a nonuniform electric field are first proposed as a special class of "biomarkers," which can be used to distinguish, sort and separate cancer cells. The aim is to develop a new high-throughput, label-free nondestructive cell separation method for the application of the early diagnosis and the personalized treatment of B-lymphoma patients. The method includes: to establish a combined electrokinetics and hydrodynamics model to achieve massively parallel manipulation of single cells with high precision; study on the label-free nondestructive separation mechanism of tumor cells and blood red cells (RBCs), and optimization of the gradient electric field; to establish an experimental platform integrating the ODEP chip with the PDMS microchannels, for discriminating and sorting the B
