中文摘要: 肝癌是我国常见恶性肿瘤,目前除手术切除及化疗外,尚无理想的治疗方法。声动力疗法是一种新兴的肿瘤治疗方法,课题组前期研究发现低频超声激活声敏剂可以有效杀伤肿瘤细胞(Ultrasonics, 2011; Ultrason Sonochem, 2008)。在此基础上我们首次提出声动力复合脂质体(专利号:201210171178.3),应用新型纳米技术,将声敏剂与传统化疗药物组成载药系统。利用脂质体的类脂结构辅助声敏剂二氢卟吩更好地聚集于肿瘤组织,通过超声波激活积聚在肿瘤细胞内的声敏剂,继而产生一系列的生物化学反应,使复合脂质体化疗具有靶向性,双重杀伤肿瘤细胞,达到协同增效作用。本课题拟制备表阿霉素-二氢卟吩复合脂质体,HPLC测定脂质体的包封率和渗透率。通过离体和在体实验,验证复合脂质体对肝癌的靶向治疗作用,并从分子、细胞、组织和整体动物水平多个层面研究其抗肝癌作用的确切机制。

英文摘要: Liver cancer is a common malignant tumor in our country. At present, there is no ideal treatment except surgery and chemotherapy. Sonodynamic therapy (SDT) was a new type of anti-cancer method. We fristly found that sonosensitizers could effectively kill tumor cells with low frequency ultrasonic activation (Ultrasonics, 2011; Ultrason Sonochem, 2008). Sonosensitizer-liposomal chemotherapeutic complex (No.201210171178.3) was firstly raised based on previous studies. Sonosensitizers and traditional chemotherapeutic drugs were combined to a drug delivery system by application of the new nano technology. Use the lipid structure of liposomes assisting sonosensitizer chlorin to target to tumor tissue. SDT, producing a series of chemical reactions activated by radiation to kill tumor cells, may be an optional treatment for localized tumors and improve effectiveness of the chemotherapy. This subject prepared epirubicin-chlorin liposomal-complex.The encapsulate rate and leakage rate of liposomes were determined by HPLC. The targeted effect of liver cancer was verifed through the in vivo and in vitro experiments. In addition, the mechanisms of anti-cancer were studied on levels of molecule, cell, tissue and the whole body.
