中文摘要: 单细胞内金属离子的检测和分析对于理解和揭示其在生命过程中的作用机理,具有重大的科学意义。但是,发展高灵敏、高选择性及高准确性的适用于单细胞内多组分金属离子的检测手段仍然是具有挑战性的课题。本项目拟通过制备介孔二氧化硅-贵金属复合材料体系,利用其高比表面积和规则有序的介孔孔道,创造高密度且分布均匀的SERS热点,构建灵敏度高、重复性好、稳定性佳、生物兼容的SERS基底。同时,设计和合成对目标金属离子具有高选择性的分子配体作为拉曼探针分子,构建灵敏度高且选择性好的SERS传感器。在此基础上,针对重要生理病理过程中的关键金属离子,通过对基底表面功能化修饰一系列探针分子,实现对单细胞内多组分金属离子的快速、准确、灵敏的检测和分析。本项目的实施将为从单细胞层面检测和研究具有重要生理病理意义的金属离子提供了一种切实可行的研究模式。

英文摘要: Metal ions play an important role in various biological processes, making their detection inside single cells of great scientific importance. However, it still remains challenging to establish a highly sensitive and selective analytical technique for the single-step multiplex detection of metal ions at the single-cell level. This project will develop a high-performance surface-enhanced Raman scattering (SERS) substrate based on mesoporous silica-metal composite materials. The large surface area and ordered mesostructure facilitate the formation of high-density and high-homogeneity hot-spots. Raman probe molecules will be designed and synthesized for selective recognition of target metal ions in order to further improve the detection selectivity. In addition, an ultra-sensitive platform for the single-step multiplex detection of metal ions inside single cells will be established through the surface functionalization of a range of different Raman probe molecules. The pathological role of metal ions will be further investigated using such SERS-based sensor. The overall methodology not only affords a new strategy for probing a variety of metal ions at the single-cell level but also provides an insight into the role of metal ions in biological system.
