中文摘要: 针对国内外Se对Cd解毒机理研究薄弱环节以及Se与Cd作用靶标细胞研究的空白现状,采用植物体硫饥饿阻断含硫有机物合成的实验方法,以及高硫“#28322;出实验”#65292;来研究Se与Cd的作用靶标。采用最新的EXAFS和SRXRF分析手段,研究Se和Cd在植物体内器官水平、组织水平、细胞亚细胞水平的分布定位及分子形态,重点是从细胞亚细胞水平阐明Se和Cd的分布定位、结合形态(Se-Cd结合态)以及价态转化在Cd的累积和解毒中的作用;进而再通过根系伸长试验和荧光染色实验对不同结合形态和不同价态的Se对Cd毒性拮抗作用进行验证。用硫限制植物同化试验来研究Se对Cd的解毒机理在国际上属开创性工作,本研究将为Se与Cd作用靶标的定位,Se与Cd在细胞中的结合形态的研究提供科学依据,为应用Se来拮抗重金属毒害提供创新的技术支持。

英文摘要: Studies on the detoxification mechanisms and the interaction between Se, Cd. Experiments will be carried out in plants to study the interaction between target cells, Se and Cd both with sulfur starvation, which inhibits the production of sulfur compounds, and high sulfur concentration ‘verflow experiment’ Extended X-ray absorption fine structure (EXAFS) and synchrotron radiation extended X-ray absorption fine structure (SRXRF) will be used to investigate the distribution, localization, and molecular morphology of Se and Cd in the plant’ organs, tissues, cells and sub-cells. The key points of this study are to clarify the distribution, localization, and binding forms (Se-Cd) of Se and Cd, and the effects of valance transfer on accumulation and detoxification of Cd at cell and sub-cell level. The effects of different binding forms and different valence of Se on the antagonism of Cd will be studied by root elongation and fluorescence experiments. The present research project represents a pioneering work in the world on the detoxification mechanism of Se and Cd by using sulfur-limited plant assimilation. This project will provide the scientific basis for the study of target localization of Se and Cd, clarify the binding forms of Se and Cd in cells, and innovate the application of Se antagonizing against heavy metal.
