中文摘要: 应激抵抗是近年来抑郁症研究领域的一个新视点。从应激抵抗角度入手,探索机体面临应激时的适应、保护性机制,可为抑郁症的预防、诊断和治疗提供新的依据。本课题组的前期研究发现,慢性温和应激(CMS)抑郁模型大鼠脑组织谷氨酰胺(Gln)含量与应激抵抗相关。因此,本课题拟首先对CMS大鼠进行Gln干预,通过行为学、生化学分析证明Gln干预的抗应激作用。在此基础上,采用蛋白质和代谢物的高通量分析技术,探讨Gln正向肠道免疫调节作用介导应激抵抗发生的相关分子及其代谢机制;同时,通过对谷氨酰胺-谷氨酸循环中关键分子的靶向分析,探讨Gln干预对该环路的影响。结合上述两种策略,将全面、深入了解Gln的正向肠道免疫调节功能在抑郁症应激抵抗中的作用机制。本课题将为抵抗慢性温和应激提供有效地干预措施,进而从一个全新的角度为发展预防抑郁症发生的干预措施提供理论依据。

英文摘要: Chronic stressful events are key risk factors for major depressive disorder (MDD), yet some individuals exposed to stressful events do not develop MDD. This disparity suggests the significance of resilience to deleterious stress effects. However, the underlying molecular mechanisms of stress resilience are poorly understood. In the present research,the chronic mild stress (CMS) rat model of depression was used to reveal the individual differences in stress response. Based on this model, the effects of glutamine supplementation on stress responses and the intestinal immunity would be estimated. Further, a combined proteome and metabolite-profiling analyses would be used to reveal surprising insights into the mechanism of stress resilience. In the meanwhile, a targeted analysis of glutamine-glutamate cycle would be conducted simultaneously. These findings would provide insight into the molecular mechanisms underlying stress resilience and shed light on novel therapeutic opportunities to augment stress resiliency.
