中文摘要: 家蚕核型多角体病毒是一类专一性感染家蚕的DNA病毒,是引起家蚕病害的主要病原。BmNPV在感染周期中会出现两种表型不同但遗传物质完全相同的病毒形态,即出芽病毒BV和包涵体衍生型病毒ODV。ODV通过经口感染寄主,介导昆虫个体间的传播,引发中肠原发感染,而BV负责感染细胞和组织间的水平传播,由此引起寄主的全身性感染。GP64是BV表面的重要特征囊膜蛋白之一,目前已经明确在BV感染过程中起关键作用, 但至今GP64蛋白的受体没有被鉴定。通过免疫共沉淀、病毒覆盖、蛋白质质谱鉴定等技术,鉴定BV GP64感染宿主细胞表面相应受体蛋白,进一步阐明其编码基因和分子特征。本研究的结果将为阐明病毒囊膜蛋白与受体的相互作用,病毒囊膜-细胞膜融合的分子机制等提供科学依据,从而明确病毒粒子进入寄主细胞的分子机理。阐明这些科学问题还将有助于揭示杆状病毒的入侵机制及规律,并为设计抗家蚕病毒入侵的有效药物提供新思路。

英文摘要: The Bombyx mori nucleopolyhedrovirus (BmNPV) is a specific pathogen to infect silkworm.During the baculovirus infection cycle,it produces two types of virions, budded viruses (BVs) and occlusion-derived viruses (ODVs).ODVs cause oral infection when the host ingested the viral occlusion,While BVs are responsible for systematic infection within the silkworm. GP64 is the most important envelop protein in BV and was proved to key role in BV infection, however, the receptor for GP64 on the surface of host cell has not yet been identified.In this study, by using the Pull-down immunoprecipitation,virus overlay protein blot assay and proteomics identification by mass spectrometry, we try to detect and identify this receptor. Furthermore, the structural basis responsible for the interaction between the GP64 and this receptor will be analyzed. The results from this study will provide important clues to understand the interaction between GP64 envelop proteins and receptor, as well as the fusion of viral envelop and cell membrane protein, and finally make clear the viral infection mechanism. Also,it will help us to design more effective chemicals to block the viral infection and give us novel insights on the strategy of pest control.
