中文摘要: CD147-MMPs信号通路在细胞外基质降解过程中发挥着关键作用,可直接造成牙周组织破坏。CD147-MMPs也是抑制其他多种疾病发展的关键靶点。多项研究提示中药成分胡椒素能对不同疾病的MMPs产生抑制作用。我们通过理论证据分析提出:胡椒素可通过调控CD147-MMPs信号通路抑制细胞外基质的降解来保护牙周组织。预实验结果初步证实胡椒素的牙周保护作用确切,并提示胡椒素的这种作用可能与调控CD147表达量和糖基化有关。研究内容:1.细胞试验研究胡椒素对CD147表达和糖基化调控的作用及对不同MMPs的作用。2. 胡椒素对炎细胞趋化性,破骨细胞增殖融合、分化和细胞骨架改建的作用,以及胡椒素对炎细胞和破骨细胞表达CD147的影响。3.胡椒素调控CD147糖链和糖基化的分子机制探索。4.动物实验体内研究胡椒素对牙周炎的相关保护作用及机制。

英文摘要: CD147-MMPs signaling pathway plays a key role in extracellular matrix degradation. It can directly cause the destruction of periodontium. CD147-MMPs signaling pathway is also the key drug target of many other diseases. Studies suggest that traditional Chinese medicine composition piperine can produce inhibitory effects on MMPs. Our theory: piperine inhibit the degradation of ECM through regulating CD147-MMPs signaling pathways. Preliminary experiment results confirmed that piperine showed exact protection, and this effect may be related to regulating the expression and glycosylation of CD147. Research contents: 1. Cells studied research that the effect of piperine on the expression and glycosylation of CD147 function and follow-up role of different MMPs. 2. Effects of piperine on inflammation cell chemotaxis, osteoclast proliferation, differentiation and the role of cytoskeleton alterations, and the regulation of CD147. 3. Piperine regulate CD147 carbohydrate chain and molecular mechanism of glycosylation. 4. Animal experiment in vivo study, relevant protective effect and mechanism of piperine on periodontitis.
