中文摘要: 舌癌的发病率位居头颈部恶性肿瘤之首,而舌癌Ⅰ期的早期诊断病例仅占10.9%~25.4%。然而,现今对舌癌的早期诊断多以有创、定性、分辨率低或对人体有害的检查方法。本项目拟将我们前期研究中的光学相干层析成像(能够实现生物组织浅表层次结构的在体显微成像,分辨率0.5-15μm)与激光共聚焦显微拉曼光谱技术(能从生化指纹方面区分不同物质结构的技术)对舌癌进行早期检测,观察舌癌的浸润深度,深层次显微变化,细胞内的蛋白质、脂类、碳水化合物和核酸等主要物质组成在结构、构象和数量上的变化,使其从形态学到分子水平上对舌癌细胞和动物模型的光学特性变化进行研究。发展一种舌癌早期无创的快速光学诊断技术。该研究有望实现舌癌诊断结果具有客观性、重复性和可比性。

英文摘要: The tongue cancer is the first on the probability of developing cancer, but early diagnosis cases of Ⅰstage of tongue cancer is only 10.9%~25.4%. But, at present, there is only invasive,qualitative diagnosis,low spatial resolution or harmful for human. We will use optical coherence tomography (which can micro-imaging on surface layer of biological tissues, the spatial resolution: 0.5-15μm) in combined with micro-Raman spectroscopy (which can distinguish cancer tissue from normal tissue on biochemistry fingerprint) to research on early tongue cancer for observing the infiltration depth,micro-change of deep layers,and the main substance of intracellular proteins, lipids, carbohydrates, and nucleic acid changes in the structure, conformation and quantity. We will research on the changement in the optical properties of tongue cancer cell and animal models from morphology to the molecular level. In order to achieve objectivity, reproducibility and comparability to the results of tongue cancer diagnosis, we will develope a tongue cancer early rapid noninvasive optical diagnostic techniques.
