中文摘要: 穴位作为针刺刺激信息的初始应答部位,对针效的发挥起着至关重要的作用。穴位通过产生各种化学物质,引起神经、体液系统的应答,因而对穴位微环境中各种效应化学物质的分析将大大推进针刺穴位效应的研究。受限于现有分析手段的低灵敏性和损伤性,针刺穴位效应物质的分析仍然困难重重,这极大地阻碍了针刺穴位效应的研究。生物传感技术具有高灵敏度、高选择性的特点,在生物分析中表现出广阔的应用前景。在此背景下,本项目拟以针灸针为基体,以特殊设计的核酸、多肽等为识别分子,构建针灸针传感界面,并结合酶辅助的信号放大策略,发展能够灵敏无损伤分析穴位微环境中化学物质的有效方法,为针刺穴位效应的研究提供新手段,为针刺作用机制和经穴特异性研究提供重要的技术支持。

英文摘要: Acupoints, the local initial sites of signals of acupuncture stimulation, play an important role in producing clinical therapeutic effects. By releasing chemicals, acupoints initiate the response of nervous system and body fluid system. Thus, the analysis of the chemicals in the regional micro-environment will greatly promote the research on the effect of acupuncture on acupoints. While considering the poor sensitivity and invasiveness of the existing research methods, new methods are in great urgency. Biosensing technology is of high sensitivity and high selectivity, which has been widely applied in biological analysis. In this project, biosensing technology is employed to study effects of acupuncture on acupoints by modifying nucleic acids and peptides as recognition elements on needles to sense chemicals non-invasively. Meanwhile, enzyme-assisted signal amplification strategy is also introduced to improve the sensitivity of biosensing needle. Thus the analysis of chemicals in the regional micro-environment can be achieved sensitively and non-invasively, which may hold great promise in the research of the acupuncture mechanisms and the specificity of acupoints.
