中文摘要: 本研究拟利用人类基因组高通量蛋白芯片技术初筛并分析阿尔茨海默病(Alzheimer's disease, AD)患者血清、脑脊液差异表达蛋白,并利用低密度蛋白芯片、western blot方法对AD各期、治疗前后、各型痴呆、其他神经变性疾病进行大样本验证,拟最终筛选出2-3个AD诊治特异的蛋白标志物,预计其中1-2个对AD临床前期、轻度认知障碍期具有早期诊断价值,1-2个对AD的鉴别诊断、疗效评价或预后具有重要价值,从而为AD的早期筛选、诊断与治疗提供依据。通过分析上述蛋白标志物的结构、亚细胞及组织定位,筛选鉴定与标志物相互作用的蛋白质,并系统研究其对Aβ、BACE1、tau蛋白表达及学习记忆能力的影响,从而初步阐明上述AD蛋白标志物的生物学功能,为AD形成、病程进展机制的研究提供下游靶标,为AD治疗靶向药物的研发提供靶点。

英文摘要: This study is firstly to screen the proteins expressed differently in the serum & CSF of Alzerimer's disease (AD)by proteome microarrays.Then,2-3 novel-specific protein biomarkers of AD will be got from the protein candidate through analyzing their levels in the specimen of all kinds of AD patients as well as other neurotics by small chip or western blot.One or two of the validated biomarkers wil be further converted to the western bolt-based assay to demonstrate their utility for early diagnosis,one or two will be used to identify or analyze the treatment effect of AD.Morever, we will utilize the current methods/technologies to study the functions of the biomarkers ,including screening and identifying differential proteins,protein-protein interactions,protein subcellular localization, and bioinformatics etc, which could further enhance our reaserch from pathology aspect.
