中文摘要: 铁死亡作为一种非凋亡细胞死亡形式,主要表现为铁沉积、线粒体膜结构的改变、胞质和脂质中ROS的增加。抑制铁死亡的发生,则可有效的预防机体发生神经退变。线粒体铁蛋白(MtFt),是一种新发现的铁蛋白,不仅能够结合铁,而且还具有亚铁氧化酶活性。我们前期的研究结果证实,MtFt对6-OHDA和Aβ造成的神经细胞损伤具有明显的保护作用,其主要机制是通过调节铁在细胞内再分配,降低自由铁池水平,从而抑制Fenton反应,减少ROS,降低氧化应激的损伤。然而调节MtFt的表达是否能够缓解神经细胞的铁死亡,至今未见报导。本研究利用过表达MtFt的SH-SY5Y神经细胞和MtFt-/-的果蝇为研究对象,阐明MtFt在铁死亡的过程中的作用及其分子机制,为全面了解MtFt的功能提供更为充足的理论基础。

英文摘要: Ferroptosis, as the nonapoptosis form of cell death, shows iron deposit, mitochondrial membrane structure changed, the increased ROS in cytoplasmic and lipid. The inhibition of ferroptosis may protect organisms from neurodegeneration. Mitochondrial ferritin (MtFt), a new discovered ferritin, is not only can combined with iron atoms, but also has ferrous oxidase activity. Our researchs confirmed that MtFt can apparently attenuate 6-OHDA and Aβ induced nerve cell damage. The protective role of MtFt is mainly through the regulation of iron in intracellular redistribution, causing iron deficiency in the cytoplasm, thus inhibiting Fenton reaction, and then reduce the levels of ROS, reduce oxidative stress damage. But whether MtFt can regulate Erastin-induced cell death has not been reported up till now. In this research, we use the MtFt-SH-SY5Y nerve cells and MtFt -/- drosophila try to clarify that in the process cell death MtFt has certain protctive effect and figure out the mechanism, the result will provide some new theoretical knowledge to fully understand the function of the MtFt.
