中文摘要: 金属钽用于骨内植入物的研究取得了重大进展,可以弥补现应用最广泛的骨内植入物金属钛在耐腐蚀性、骨诱导活性和抗菌性等方面的不足,我们的前期研究也显示用离子注入技术的钽涂层具有更为优越的促骨髓间充质干细胞增殖能力。然而将金属钽应用于牙种植体仍然存在着一些困难。本项目创新性地利用离子注入技术,以临床常用的酸蚀喷砂(SLA)的钛种植体为基底,构建新型的钽涂层修饰的SLA钛种植体,研究其生物相容性、抗腐蚀性、骨诱导性及抗菌性等生物活性。研究将从整体、细胞和分子生物学不同层面上,阐明其诱导骨组织再生和发挥抗菌活性的作用途径及其机制,验证integrin-linked kinase/β-catenin和Wnt /β-catenin通路中的关键功能蛋白在诱导骨再生中的作用。项目最终试图解决金属钽修饰钛种植体的制备技术、生物学效应、骨整合能力、抗菌效果及相关的分子机制等问题,为开发新一代牙种植体提供科学依据。

英文摘要: Research of tantalum as bone implantation has made great progress, it could make up for the shortcomings of widely used tatanium, such as poor corrosion resistance, lack of bone induction activity and weak antimicrobial properties. However, the application of tantalum in dental implantology still faces some difficulties. We use ion injection technology to build a tantalum modified SLA implants on the basis of SLA implants. We try to assess the biocompatibility, corrosion resistance, bone induction and antimicrobial properties of the new materials. Our previous study has shown that the tantalum modified SLA implants could induce more rBMSCs proliferation. In addition, we try to clarify the pathways and mechanism of the bone induction activity and antimicrobial properties on overall, cell and molecular levels and validate the role of key protein in integrin-linked kinase/β-catenin and Wnt /β-catenin signal pathways. We try to assess the tantalum modified implants preparation technology, biological effect, osseointegration ability and antibacterial effect. We try to comprehensively evaluate the potential of clinical application, to fill the blank of the present research and to provide the scientific basis for a new generation of dental implants.
