中文摘要: 靶向治疗和药物可控释放是化学科学、生物材料、药物科学领域研究工作者共同面临的难题。一个重要的目标即是实现药物定时、定量、定位的控制释放,提高药物的长期使用效率即长效性。本项目拟设计大分子链中含超声波力化学响应基团的系列新型动态聚合物,表征产物结构。在载药无机纳米介孔二氧化硅存在下,实现动态聚合物与无机纳米粒子的接枝或包覆,制备稳定的无机纳米介孔二氧化硅-动态聚合物复合体系。考察聚焦超声参数对介孔二氧化硅表面动态聚合物分子链结构的影响,研究聚焦超声定位降解的空化机制以及定位降解特征。研究超声场对动态化学键分解和形成平衡过程影响,建立反应动力学模型。研究复合粒子装载药物或荧光分子的超声释放动力学。实现聚焦超声波远程控制载药无机纳米介孔二氧化硅-动态聚合物复合粒子的药物释放,建立超声波调控的药物释放物理和化学分子机制。评价聚焦超声控制药物释放的生物安全性和药效.

英文摘要: Targeting therapy and controlled release of drug remain big challenges for the researcher in the field of chemistry, biomaterials and pharmaceutical science. One important aim is to realize the controlled release at the disease site, on time and at a required amount and to enhance the long term effect of the drug. This project is to design and synthesize the novel ultrasound responsive dynamic polymer by introducing mechanophore group into the molecular chain, and to characterize the polymer structure. Furthermore, the dynamic polymer was encapsulated or grafted onto the surface of the mesoporous silica to obtain the hybrids of mesoporous silica and dynamic polymer.The effect of high intensity focused ultrasound (HIFU) parameters on the structure of dynamic polymer will be examined and the behavior and mechanism of HIFU-induced site specific degradation of polymer will be examined. The decomposition and recombination of dynamic chemical bond under the ultrasound field will be investigated, the reaction kinetics model will be established.In addition, the drug or fluorescent chemicals will be introduced to the hybrids of mesoporous silica and dynamic polymer. The kinetics and mechanism of drug release from the composite particlers under the stimuli of HIFU will be studied. Eventually,the remote HIFU controlled on-off drug release will be realized. Also, the safety and efficacy of HIFU-controlled drug release from dynamic copolymer-mesoporous silica will be evaluated through the in-vitro cell cytotoxicity experiments.
