中文摘要: 苯乙醇胺A(PEAA)是养殖中新出现的β-受体激动剂类违禁添加物,其体内代谢规律及对生理过程的影响还不清楚。代谢组学可以同时阐明外源物代谢过程及其内源代谢响应,非常适合兽药(或化学污染物)的体内代谢和残留研究。本项目将应用基于超高效液相色谱-四级杆-飞行时间质谱的代谢组学方法,从整体水平上阐明动物摄入PEAA后的代谢组轮廓变化,借助多元数据分析模型(PCA和OPLS-DA),筛选PEAA的体内代谢产物和生物标志物,揭示PEAA的体内代谢规律及其对动物内源代谢过程的影响。同时,通过研究PEAA及其代谢产物的消除规律,确定PEAA在猪尿中的残留标示物。本研究将有助于系统认识PEAA在食品动物体内的代谢和残留机制,为残留监控和风险评估提供理论基础和科学依据。

英文摘要: Phenylethanolamine A (PEAA) is a new β-agonist compound, and its illegal use for growth promoting purpose has been observed . Hower, its metabolism and physiological effects in animal are still unclear. Metabolomics is a systemic approach to study xenobiotic metabolite profile and endogenous metabolic response, which is suited for application to study of veterinary drug metabolism and residues in animal. In this project, based on ultra-liquid chromatography coupled to quadrupoles-time-of-flight mass spectrometry, a metabolomic approach will be developed to globally study PEAA metabolism and its endogenous metabolic respones in rat and pig. By the means of multivariate data analysis, PEAA in vivo metabolites and potential bio-marker will be screened and identified. Meanwhile, according to depletion study of PEAA and its metabolites, residue marker in pig urine will be determined. This study can contribute to understanding in vivo metabolism and residue kinetics of PEAA in food-producing animals, and provide valuable scientific basis for residue control and risk evaluation of PEAA.
