中文摘要: 微小反向转座子(MITEs)在植物中普遍存在,但对其研究并不多,尤其是用实验方法获得MITEs信息的报道极少。同时,还没有对植物基因组中MITEs的系统研究,其在植物基因组中的作用价值也易被忽略。本项目拟系统鉴定已经测序的植物基因组中MITEs序列,分析MITEs序列特点以及它们在基因组中与基因分布的关系,了解MITEs的扩增方式,MITEs对植物基因表达的影响,建立一个植物MITEs数据库,为植物中MITEs的研究提供必要的信息。拟通过生物信息学方法鉴定活跃MITEs,并通过实验方法验证其转座功能。活跃的MITEs意味着它可以跳动并整合到基因组其他位置,因而可以成为遗传工具的一种。将验证的活跃MITE转化番茄,评估使用活跃MITEs系统构建突变体库的可行性,为突变体库创建提供新的手段。

英文摘要: Miniature inverted-repeat transposable elements (MITEs) are prevalent in eukaryotic species including plants. There have been few studies on MITEs, and only little information on MITEs was experimentally acquired. There is still no report on systemic study of MITEs , and the value of MITEs in plant genomes was neglected. The project will genome-wide de novo identify MITEs in all sequenced plant genomes and analyze MITEs distribution, amplification and their regulation on genes. To provide more information about MITE research, we will build a plant MITE database. According to the MITE sequence information, we will identify active MITEs from all plant genomes. Active MITEs can be integrated into or removed from genome sequences, so they can become a genetic tool for studies on plants. Here, we will identify active MITEs and their transposase, then transform them into tomato. Their transposition in tomato will be investigated and the feasibility of constructing a mutant bank using this system will be evaluated.
