中文摘要: CCC1家族蛋白是一类跨膜转运铁或锰离子的膜蛋白。该家族蛋白通常有250-400个氨基酸,5次跨膜螺旋。目前国际上尚未解析此家族蛋白的高分辨率结构。 本研究计划解析古细菌CCC1家族铁离子转运蛋白的高分辨率三维结构,阐明铁离子跨膜转运的机理。 研究内容包括用大肠杆菌异源表达目的蛋白,用镍柱亲和、凝胶阻滞等层析方法纯化蛋白,再用多种商业化结晶试剂盒筛选晶体。晶体初筛时会使用传统的低去垢剂浓度低脂质浓度的二型膜蛋白晶体生长法,最新发展的高去垢剂浓度高脂质浓度的HiLiDe一型膜蛋白晶体生长法以及LCP脂立方相膜蛋白晶体生长方法。 获得初始晶体后,对去垢剂、pH值、沉淀剂及盐浓度等进行精细优化以得到最优晶体,在同步辐射光源测试晶体衍射能力,收集衍射数据。同时,通过浸泡重金属化合物和硒代甲硫氨酸修饰目的蛋白的方法获取相位信息进而解析结构。基于结构分析和活性实验结果,使铁离子跨膜转运的机理得到阐明。

英文摘要: CCC1 family contains membrane proteins that transport iron or manganese ions across the membrane. They usually have 250-400 residues and 5 transmembrane helices. No high resolution protein structures have been determined for this family so far. We plan to study the 3D structure of archaea CCC1 family iron transporters and elucidate the iron transporting mechanism. In this study, the recombinant target protein will be expressed in E. coli, purified with nickel affinity, gel filtration and other chromatography methods. Crystal screening will be performed against a variety of commercial screening kits. During initial crystal screening, the conventional low detergent and lipid concentration method aiming for type II membrane protein crystals will be used, as well as the newly developed HiLiDe method using high lipid and detergent concentration for type I membrane protein crystal growth, and the LCP (Lipid Cubic Phase) method which is also for type I membrane protein crystal growth. Once the initial crystals are obtained, crystal optimization on detergents, pH, precipitant and ion type and concentration will be performed.
