中文摘要: 研制和开发新型纳米药物载体对解决靶向药物传输系统的准确定位以及药物的控释有着重要的科学意义和实用价值。本项目针对当前有机高分子聚合物和无机多孔材料等药物载体的不足,以药物搭载-释放为功能导向,利用不同尺度的有机功能基块和无机金属离子(金属簇),通过化学自组装技术构筑具有微孔、介孔等纳米孔道结构的新型药物搭载材料- - 纳米多孔金属有机骨架材料;进而,在各类表征的基础上,以三峡地区中草药活性成分的搭载-控释为探针,从理论和实验相结合的角度探索纳米多孔金属有机骨架材料的合成条件、结构特征、物化性质及其与药物搭载-控释的内在关联,揭示其机理,总结其中规律,研制高搭载量且控释性能优越的纳米多孔金属有机骨架材料,推进其在药物搭载及控释方面的研究进程,丰富超分子化学、配位化学、材料化学和药物化学的研究内容。

英文摘要: It has important scientific significance and practical value for research of novel nanometer drug carriers with accurate positioning and controlled release in targeting nano-drug-delivery system. The project is to construct novel nano-porous metal-organic frameworks with micro or mesopores which self-assembled from different organic ligands and metal ions(or metal clusters) to solve the disadvantage of organic polymer and inorganic porous materials, as the guidance of drug delivery and controlled release. And then, in the basis of all kinds of characterization, take the drug release of Chinese herbal medicine active ingredients in the three gorges region as a probe, we will study from theoretical and experimental aspects to explore the synthesis conditaions, structural features, physical properties, controlled-release effects and reveal the internal relation, reaction mechanism and find high-carried nano-porous metal-organic framework materials. According to the above researches, it not only promote the research process of drug release of metal-organic frameworks, but also enrich the contents of supramolecular chemistry, coordination chemistry, materials chemistry and pharmaceutical chemistry.
