中文摘要: 水凝胶是一种优异的药物载体材料,但在输送疏水药物和基因方面具有很大的局限性。基于α-环糊精和聚乙二醇(PEG)的主客体相互作用形成的超分子水凝胶,因其具有制备简单、条件温和、性质易于调控、环境响应等优点,被广泛用作可注射药物载体进行研究。此外,单分子胶束作为一种新型的药物/基因载体,具有各种优异的性能而受到广泛关注。由疏水链段修饰的超支化聚甘油衍生物是一种优异的单分子胶束,被用于药物/基因载体。为此,提出本课题的研究思路:以外周由PEG修饰的超支化聚甘油单分子胶束为基材,通过其外周PEG链段与α-环糊精的包合作用,构建一种可注射的超分子水凝胶,其中,超支化聚甘油单分子胶束的疏水性内核能够用于负载疏水药物;在超支化聚甘油的羟基上接枝聚赖氨酸(PLL)分子,可用于包裹基因分子,因此该体系具有疏水药物和基因联合给药的优势。而且,该水凝胶体系还具有pH和温度响应性能,能够用于药物和基因的智能释放。

英文摘要: Hydrogel is an excellent material for drug delivery, but has limitation in delivering genes and water-insoluble drugs. Supramolecular hydrogels, based on the host-guest interaction between α-cyclodextrin and polyethylene glycol (PEG), are widely investigated as injectable drug carriers, due to their simple and mild preparation condition, tunable properties, and environmental sensitivity. In addition, unimolecular micelles as a kind of novel drug/gene carriers have good properties and attract much attention. Polyglycerols-based unimolecular micelles bearing hydrophobic segments and polylysine (PLL) chains, have been studied for drug/gene delivery. Based on these studies, we put forward the idea of this project: polyglycerols-based unimolecular micelles with PEG periphery interact with α-cyclodextrin to form injectable hydrogels. Within the hydrogels, the hydrophobic inner-cores of the micelles can be used to load water-insoluble drugs. By conjugating PLL chains to the polyglycerols, the hydrogels can also be used for gene delivery. Therefore, the supramolecular hydrogels have the advantage of co-delivery of genes and water-insoluble drugs. Moreover, the hydrogels have pH- and temperature-sensitivity, which can be used for smart release of drugs and genes.
