中文摘要: 稳定同位素化学标记技术已被大量应用于生物体系的定量蛋白质组学研究。基于二级质谱的定量蛋白质组学方法不会增加一级质谱复杂度,且具有较高的灵敏度,但该类方法仍存在不足,如低端报告离子或序列信息离子信号抑制,影响了定量的准确度和覆盖率。针对目前存在的问题,本项目合成了同位素标记的异硫氰酸酯类标记试剂,该类试剂标记的多肽能够发生Edman 降级式碰撞裂解,产生较高丰度报告离子,同时不丢失测序碎片信息,且具有更好的质谱兼容性和灵敏度。基于此类试剂建立了新型等重肽末端标记策略 (IPTL) 和新型等重标签 策略(IT),用于提高相对定量的精确度、准确度和覆盖率。利用所建立的方法对喉癌细胞与正常喉细胞蛋白质组进行分析,从喉癌细胞中筛选出潜在的生物标志物,并对差异蛋白进行生物信息学分析,以期为喉癌早期诊断标志物的研究提供参考,为进一步从分子水平阐明喉癌的发生发展机理提供详实数据。

英文摘要: The chemical labeling measuring techniques based on stable isotope followed by mass spectrometric analysis have been widely used in quantitative proteomics studies. The method based on MS/MS for the proteomics studies has high sensitivity and exhibits more excellent properties than MS method that will increase the complexity of data analysis, but MS/MS method still has some shortcomings, such as the signal suppression of low weight reporter ion or sequence information ion, and affects the accuracy and coverage for the data analysis. In view of these problems, we synthesized a series of isotope labeled isothiocyanate ester reagents. The labeled peptides by these reagents easily give the Edman-Type cleavage, and produce high abundance reporter-ions, without loss of the information of sequence fragments. At the same time, these reagents have better MS compatibility and sensitivity. In this project, we will develop two methods based on these reagents, namely Isobaric Peptide Termini Labeling (IPTL) strategy and Isobaric Tag (IT) strategy. Two new methods can improve the precision, accuracy and coverage in protein sample analysis. The established methods will be employed to analyze the proteome of laryngeal cancer cells and normal laryngeal cells, and screen out a lot of potential biomarkers. The biological function
